Only the tutorial can be played off disc. An update is required in order to play offline private matches.
Even with the update, offline private matches are extremely limited. You cannot earn any XP and will be stuck with the default loadouts.
If you are playing as the humans offline, there is no AI bot for the Predator meaning the only threat will be the enemy soldiers. When playing as the Predator offline, the Soldier AI is extremely basic and will forever be stuck searching for you if you ever get spotted by them.
